Job Description

This role will be based at City St George's University of London Tooting campus. The vision for the project is to deliver a novel solution to enable Mycobacterium tuberculosis DNA extraction from samples without the need for expensive or mechanical devices.


The project requires familiarity with CL2/CL3 pathogen manipulation and handling, molecular cloning and preferably experience with working with bacteriophage.


Main activities will include:

Development of a straightforward and inexpensive Mycobacterium tuberculosis (Mtb) lysis solution, comprising pore-forming short cationic peptides and a mycobacteriophage that together will be able to induce both host and mycobacterial cell membrane disruption, resulting in Mtb DNA release into a stable buffer compatible with downstream NAAT processing. Assess the robustness and sensitivity using quantitative microbiological molecular methods for the lysis solution to process multiple sample types, concentrating on samples of tongue swabs, exhaled breath condensate and sputum. Assess techniques to maximize overall sensitivity by enrichment of Mtb bacilli through inclusion of a mycobacteriophage capture step prior to commencement of lysis plus cloning and application of various lytic agents.


Key attributes of the successful applicant:

BSc & PhD in biomedical sciences (or closely related subject) Knowledge of CL3 handing. Knowledge of cloning and purification of recombinant proteins. Knowledge of bacteriophage handling and biology. Knowledge of techniques enabling molecular diagnostics. Strong communication skills with good written and verbal English, with ability to develop strong, effective relationships and work effectively with collaborators Ability to write laboratory reports and academic manuscripts Experimental research mind-set. Ability and flexibility to meet deadlines and tight time scales. Enjoys responsibility and solving unexpected problems with commitment to continuous improvement.
